Tyrosine-tubulin Staining

Tyrosine-tubulin staining is an immunofluorescence method that labels tyrosinated α-tubulin, a post-translationally modified form associated with dynamic microtubules in neurons and other cells. The method uses antibodies that recognize the exposed C-terminal tyrosine of α-tubulin, allowing researchers to visualize microtubule populations undergoing cycles of polymerization, depolymerization, and detyrosination. In neuroscience, this staining helps map microtubule organization in axons, dendrites, and developing neurites, providing insight into neuronal polarity, intracellular transport, and cytoskeletal remodeling. Combined with microscopy and complementary tubulin markers, it can reveal changes in microtubule stability during development, injury, or disease.

Optimizing Tubulin Yield from Porcine Brain Tissue

This protocol describes a technique for the high-yield isolation of tubulin from the porcine brain optimized for small-scale instrumentation.
