Curli Overproduction

Curli overproduction is the engineered increase of curli, extracellular protein fibers that form part of the bacterial biofilm matrix, to enhance a cell’s ability to display or assemble functional materials. In bacteria such as Escherichia coli, this process typically involves increasing expression of the csg gene system, which produces the CsgA subunit and its assembly factors; CsgA is exported and polymerizes into amyloid fibers outside the cell. In bioengineering, curli overproduction supports the fabrication of living materials, surface coatings, biosensors, and protein-based scaffolds by presenting functional peptides on robust fibers. Controlling expression is essential for balancing material production with cellular growth and stability.
