Floxed Stop Codon

A floxed stop codon is a genetic control element in which a transcriptional stop sequence is flanked by loxP sites, allowing gene expression to be activated conditionally. In the presence of Cre recombinase, site-specific recombination excises the stop cassette, placing the downstream gene under the control of its promoter; without Cre, transcription remains blocked. This design enables precise spatial, temporal, or inducible control of transgenes in engineered cells and organisms. In bioengineering, floxed stop codons support conditional gene activation, lineage tracing, functional studies, and the development of experimental models in which genetic effects can be isolated within selected tissues or treatment conditions.
