Integrin Engagement

Integrin engagement is the process by which cell-surface integrin receptors bind extracellular-matrix ligands or engineered adhesive cues, linking cells to their surrounding environment. Ligand binding can trigger integrin activation and clustering, while connections to the actin cytoskeleton and focal-adhesion proteins transmit biochemical and mechanical signals that regulate adhesion, spreading, migration, survival, and differentiation. In bioengineering, controlling integrin engagement through biomaterial chemistry, ligand presentation, substrate stiffness, and surface patterning helps guide cell behavior in tissue scaffolds, organ-on-chip systems, and regenerative therapies. Measuring these responses also clarifies how cells sense and remodel their physical surroundings.

Integrins

Animal and protozoan cells do not have cell walls to help maintain shape and provide structural stability. Instead, these eukaryotic cells secrete a sticky mass of carbohydrates and proteins into the spaces between adjacent cells. This network of proteins and molecules is called an extracellular matrix or ECM. Some ECM proteins assemble into a basement membrane to which the remaining components adhere. Proteoglycans typically form the bulk of the ECM while fibrous proteins, like collagen,...

Activation of Integrins

Integrins bind ligands and transmit information from outside the cell to inside or vice-versa through an "outside-in signaling" or "inside-out signaling." In "outside-in signaling," external factors in the extracellular space bind to exposed ligand binding sites on integrins. This causes the inactive protein to undergo a conformational change to become active. Integrins are often clustered on the cell membrane. Repetitive and regularly spaced ligand binding events provide an effective stimulus.

Measuring Engagement of Spectators of Social Digital Games

2021

We propose a methodology that enables measuring engagement of spectators in a social digital game combining physiological and self-reported data. As this digital game involves a group of freely moving people, the experience is filmed using a synchronizing technique that links physiological data with events in the game.

Evaluation of Immunomodulatory Activity of a Virally Encoded Bispecific T-Cell Engager

Source: Heidbuechel, J. P., et al. Paramyxoviruses for Tumor-targeted Immunomodulation: Design and Evaluation Ex Vivo. J. Vis. Exp. (2019).This video demonstrates a cell-based assay to evaluate the target-specific immunomodulatory activity of a virally encoded bispecific T-cell engager (BTE). The BTE links tumor cells expressing a target receptor with T cells, triggering tumor lysis and enzyme release. A colorimetric readout, based on substrate conversion, indicates T-cell activation. Higher...
