Minimum Media

Minimum media are chemically defined growth formulations that provide only the essential nutrients required for microorganisms or cells to survive and reproduce, making them valuable for controlled bioengineering experiments. They typically combine a carbon and energy source with inorganic salts, nitrogen, and selected trace nutrients; cells must synthesize omitted metabolites through their own biosynthetic pathways. By reducing undefined components, minimum media help researchers isolate nutrient requirements, evaluate metabolic pathways, and assess engineered strains under reproducible conditions. These formulations support microbial cultivation, auxotroph selection, metabolic engineering, and production studies, while revealing how nutrient availability influences growth, adaptation, and cellular performance.

Determining the Minimum Inhibitory Concentration of Enrofloxacin Against Avian Pathogenic E. coli

2026

Begin with a suspension of avian pathogenic E. coli or APEC in growth media.APEC is a Gram-negative bacterium that infects poultry by entering through the respiratory tract. After breaching the mucosal layer, APEC adhere to epithelial cells, multiply locally, and disrupt the epithelial barrier to enter the bloodstream. The bacteria then spread to multiple organs, causing systemic infection.Add an equal volume of the bacterial suspension to each well of a multi-well plate containing increasing...

A Microtiter Plate Method to Assess the Minimum Inhibitory Concentration of an Antibiotic

2025

This video demonstrates a microtiter plate-based method for determining the Minimum Inhibitory Concentration (MIC) of test antibiotics against bacteria. Upon adding a suspension of the bacterial pathogen Pseudomonas aeruginosa to a microtiter plate containing serial dilutions of a test antibiotic, and an inhibitor of the production of pyomelanin by the bacteria, the optical density of the wells is measured to compute the minimum inhibitory concentration of the antibiotic, and the impact of the...

Local Maximum and Minimum Values

2026

In multivariable calculus, a function of two variables can exhibit local maximum or minimum values at certain points on its surface. A local maximum occurs when the function's value at a point is greater than at all nearby points, while a local minimum occurs when the function’s value is less than at all nearby locations. These points are referred to as local extrema and are of central importance in optimization problems.Local extrema are found at critical points, where the surface becomes...

2017

We present an apparatus based on hot-wire ignition in a pressurized enclosure and an associated methodology to measure the minimum pressure required to induce sustained combustion in water-based emulsion explosives. This method improves product characterization to allow one to use them more safely during pumping and mixing operations.
