Murine Fat Tissue

Murine fat tissue is adipose tissue obtained from mice, a tractable mammalian model for studying energy storage, endocrine signaling, inflammation, and tissue remodeling. Its white, brown, and inducible beige adipocytes respond to nutritional and thermal conditions through lipid storage or mitochondrial thermogenesis, while extracellular matrix, blood vessels, and immune cells shape the tissue microenvironment. In bioengineering, murine fat tissue supports investigations of adipogenesis, biomaterial design, vascularization, and engineered adipose grafts, with tissue-derived cells or matrix used to evaluate how scaffolds and culture conditions influence regeneration. These models help connect material properties and cellular behavior to metabolic function, therapeutic testing, and translational adipose-tissue constructs.

We present a protocol to isolate adipose tissue-derived microvascular fragments that represent promising vascularization units. They can be rapidly isolated, do not require in vitro processing and, thus, may be used for one-step prevascularization in different fields of tissue engineering.

An ex vivo Mouse Model for High-Fat Diet-Like Adipose Tissue Inflammation

Described herein is a protocol to isolate and analyze the infiltrating leukocytes of tissues at the maternal-fetal interface (uterus, decidua, and placenta) of mice. This protocol maintains the integrity of most cell surface markers and yields enough viable cells for downstream applications including flow cytometry analysis.
