Octanol-water Partition

Octanol-water partition is a measure of how a chemical distributes between an organic octanol phase and an aqueous water phase at equilibrium, providing an estimate of its relative hydrophobicity or lipophilicity. The partition coefficient, commonly expressed as logP, reflects the concentration ratio of the un-ionized compound in octanol versus water; for ionizable substances, pH-dependent distribution is described by logD. In bioengineering, these properties help predict membrane permeation, solubility, biodistribution, and drug release from biomaterials. Octanol-water partition data therefore supports the design and screening of pharmaceuticals, delivery systems, and other biologically active molecules.

On-Chip Octanol-Assisted Liposome Assembly for Bioengineering

The present protocol describes octanol-assisted liposome assembly (OLA), a microfluidic technique to generate biocompatible liposomes. OLA produces monodispersed, micron-sized liposomes with efficient encapsulation, allowing immediate on-chip experimentation. This protocol is anticipated to be particularly suitable for synthetic biology and synthetic cell research.

A technique to probe the lipid raft partitioning of fluorescent proteins at the plasma membrane of living cells is described. It takes advantage of the disparity in diffusion times of proteins located inside or outside of lipid rafts. Acquisition can be performed dynamically in control conditions or after drug addition.
