Rat Spleen Scaffold

A rat spleen scaffold is a biomaterial derived from rat splenic tissue for studying organ structure, extracellular matrix function, and tissue regeneration in bioengineering. Typically produced through decellularization, it removes cellular material while preserving key extracellular matrix components and aspects of the spleen’s three-dimensional architecture and vascular organization. This preserved framework can support cell attachment, migration, and tissue-specific remodeling when repopulated with suitable cells under controlled culture conditions. Rat spleen scaffolds provide experimental models for investigating splenic repair, immune-system engineering, biomaterial-host interactions, and strategies for constructing functional tissue, while enabling researchers to evaluate scaffold composition, biocompatibility, and regenerative potential.

This protocol introduces a method to develop a scaffold using decellularized rat kidneys. The protocol includes decellularization and recellularization processes to confirm bioavailability. Decellularization is performed using Triton X-100 and sodium dodecyl sulfate.

A Mouse Model of Vascularized Heterotopic Spleen Transplantation

2025

The video demonstrates a surgical method for generating a mouse model of vascularized heterotopic spleen transplantation. The abdominal cavity of an anesthetized donor mouse is surgically opened to remove the spleen. The inferior vena cava and infrarenal aorta are identified, their lumbar branches are ligated, and openings are made for the heterotopic transplantation of a spleen from a congenic donor mouse. Upon completing vascular anastomosis and restoring blood flow to the implanted spleen,...

Assessing Bacterial Burden in the Mouse Spleen for Infection Studies

(2016)This video demonstrates the process of quantifying bacterial load in the spleen of an infected mouse to assess the severity of infection. The homogenized spleen is serially diluted and plated to determine total bacterial burden. The resulting data provide insights into host-pathogen interactions within...
