Adapter Ligation

Adapter ligation is a molecular biology technique that attaches short, designed nucleic acid sequences to DNA or RNA fragments, enabling their subsequent handling and analysis. In a typical reaction, a DNA ligase forms covalent phosphodiester bonds between compatible ends of the adapters and target fragments, often after end repair or enzymatic preparation; adapters may also provide primer-binding sites, indexes, or platform-specific sequences. This process is central to constructing sequencing libraries, where ligated fragments are amplified, selected, and analyzed to identify genetic variation, measure gene expression, or characterize genomes and transcriptomes. Efficient ligation helps preserve library complexity and supports accurate downstream results.
