Calorimetric Vials

Calorimetric vials are specialized containers used to hold samples during calorimetry, allowing researchers to measure heat released or absorbed by a biological system. In a calorimetric experiment, a reaction, binding event, or metabolic process changes the temperature of the vial and its surroundings; the instrument detects this change and converts it into thermal energy using calibration and the system’s heat capacity. In biology, these vials support measurements of enzyme activity, microbial metabolism, biomolecular interactions, and cellular energy use. Careful control of sample volume, sealing, and thermal equilibration improves reproducibility and helps link heat changes to underlying biological processes.

Take serial dilutions of a bacterial culture and add aliquots into calorimetric vials containing growth medium.Load the sealed vials into the multi-well plate of an isothermal microcalorimeter to measure metabolic heat production. The device is held at a constant temperature to ensure the detected heat originates solely from bacterial metabolism. As bacteria metabolize nutrients and proliferate, they release heat that sensors detect in real time.In parallel, incubate identical vials in an...

This article presents an optimized yeast estrogen screen for quantifying ligands in Personal Care Products (PCPs) that bind estrogen receptors alpha (ERα) and/or beta (ERβ). The method incorporates two colorimetric substrate options, a six-day refrigerated incubation for use in undergraduate courses, and statistical tools for data analysis.

An Inexpensive, Scalable Behavioral Assay for Measuring Ethanol Sedation Sensitivity and Rapid Tolerance in Drosophila

Straightforward assays for measuring ethanol sensitivity and rapid tolerance in Drosophila facilitate the use of this model organism for investigating these important ethanol-related behaviors. Here, a relatively simple, scalable assay for measuring ethanol sensitivity and rapid tolerance in flies is described.

A Method to Inflict Closed Head Traumatic Brain Injury in Drosophila

REF and WEF Fly Genetics ExpandTo prepare for the experiment, be sure to obtain cultures of Drosophila melanogaster at least two weeks ahead of the planned experiment day. NOTE: Because adult females store sperm after mating, you will need this time to generate virgin females by breeding the flies. You should get two stock cultures: wildtype, that is homozygous dominant red eyed flies, and white chromosome one mutant, which are homozygous recessive white eyed flies. Purchase or create...
