Dna Polymerase Proofreading

DNA polymerase proofreading is a quality-control process that corrects errors during DNA replication, helping preserve genetic information and limit mutation. As a polymerase extends a new strand in the 5′-to-3′ direction, it detects an incorrectly paired nucleotide, pauses synthesis, and uses 3′-to-5′ exonuclease activity to remove the mismatch before resuming extension. This mechanism increases replication fidelity and helps explain how cells maintain genome stability, how mutations arise when error correction fails, and why polymerase accuracy is important in molecular biology methods that copy DNA.

This protocol describes the characterization of DNA polymerase synthesis of modified DNA through observation of changes to near-infrared fluorescently labeled DNA using gel electrophoresis and gel imaging. Acrylamide gels are used for high resolution imaging of the separation of short nucleic acids, which migrate at different rates depending on size.
