Ebola Virus

Ebola virus is an enveloped, filamentous, negative-sense single-stranded RNA virus that causes Ebola disease, a severe infection with major implications for biology, medicine, and public health. It enters susceptible cells through surface receptors, releases its RNA genome into the cytoplasm, and uses viral proteins and host machinery to produce new virions; infection can disrupt immune regulation and vascular function. Studying Ebola virus supports research on viral replication, host-pathogen interactions, molecular diagnostics, antiviral therapies, and vaccine development. These efforts improve outbreak detection and control while strengthening biosafety practices for work with high-consequence infectious agents.

Modeling The Lifecycle Of Ebola Virus Under Biosafety Level 2 Conditions With Virus-like Particles Containing Tetracistronic Minigenomes

Work with infectious Ebola viruses is restricted to biosafety level 4 laboratories. Tetracistronic minigenome-containing replication and transcription-competent virus like particles (trVLPs) represent a lifecycle modeling system that allows us to safely model multiple infectious cycles under biosafety level 2 conditions, relying exclusively on Ebola virus components.

(2014)This video demonstrates how a tetracistronic minigenome plasmid mimicking the Ebola virus genome is transfected into human cells along with helper plasmids. Inside the cells, viral proteins and luciferase are expressed, leading to the assembly and budding of non-pathogenic, replication-competent virus-like...
