Gel Disposal

Gel disposal is the safe collection, treatment, and removal of discarded laboratory gels, such as agarose or polyacrylamide gels used in biological research. It involves assessing the gel’s composition and contaminants, separating gels from electrophoresis buffers and staining solutions, and following procedures for decontamination, solid waste handling, or hazardous waste collection. Proper disposal reduces exposure to DNA-binding dyes, acrylamide residues, biological materials, and other laboratory chemicals. In biology laboratories, these practices support regulatory compliance, protect researchers and the environment, and promote responsible management of waste generated during nucleic acid and protein electrophoresis.

2023

Robert M. Rioux and Taslima A. Zaman, Pennsylvania State University, Pennsylvania, PA Users are responsible for the proper disposal of the waste generated during their work. Improper waste disposal may severely endanger public health and/or the environment. The handling of hazardous waste must be regulated from the moment of generation until its disposal at its offsite final destination facility. A waste management system must be devised before work begins on any laboratory activity. Users...

Gel Purification

2023

Gel purification is used to recover DNA fragments after electrophoretic separation. DNA recovery from an agarose gel includes three basic steps: binding, washing and eluting from a silica column. DNA is believed to bind to silica in the presence of high salt via a salt bridge. Following binding, DNA is washed of impurities and eluted under low salt conditions disrupting this interaction. This video goes through a step-by-step, generalized procedure for cutting out a band from the gel, gel...

Disposing of Accounts Receivable

Disposing of accounts receivable refers to the process of removing unpaid invoices from a company’s books, either by selling them or writing them off. This is a critical part of managing working capital and ensuring accurate financial reporting. One common approach is factoring, where a company sells its receivables to a third party at a discount. The buyer, known as a factor, provides immediate cash but keeps a portion of the receivable as a fee. While this results in a financial loss, it...

DNA Gel Electrophoresis

2023

DNA gel electrophoresis is a technique used for the detection and separation of DNA molecules. An electric field is applied to a gel matrix comprised of agarose, and within the gel, charge particles will migrate and separate based on size. The negatively charged phosphates of the DNA backbone cause DNA fragments to move toward the anode - a positively charged electrode. The video explains the mechanism by which DNA fragments are resolved on an agarose gel, and it provides a step-by-step...
