Iodine Staining

Iodine staining is a simple biological technique that uses iodine-containing solutions to reveal specific substances in cells, tissues, or biological samples. Iodine molecules interact with polysaccharides such as starch, producing a characteristic blue-black color, while glycogen may appear reddish-brown under suitable conditions. In biology, this color reaction supports qualitative tests for starch in plant tissues, food samples, and isolated cells, and can improve contrast during microscopic examination. Because the procedure is rapid and inexpensive, iodine staining is widely used in teaching laboratories and research to identify carbohydrate storage, assess tissue composition, and visualize changes associated with metabolism.
