Muscle Dissociation

Muscle dissociation is the laboratory process of separating intact muscle tissue into individual cells or small cell populations, enabling researchers to study muscle biology at cellular resolution. It typically combines enzymatic digestion, often with collagenase or related proteases that break down extracellular matrix, with gentle mechanical trituration; controlled temperature, incubation time, and handling help preserve cell viability and surface markers. The resulting suspension can support primary muscle-cell culture, isolation of satellite cells, flow cytometry, transplantation studies, and analyses of regeneration, development, or disease. As a foundational preparation method, muscle dissociation links tissue-level structure with cellular and molecular measurements.

Utility of Dissociated Intrinsic Hand Muscle Atrophy in the Diagnosis of Amyotrophic Lateral Sclerosis

Dissociated atrophy of intrinsic hand muscles, termed the split hand, appears to be a specific feature of amyotrophic lateral sclerosis (ALS). Consequently, a novel neurodiagnostic test, termed the split hand index, was developed to quantify the clinical phenomenon of the split hand, which differentiated ALS from mimic disorders.

We describe a protocol to obtain enzymatically dissociated fibers of different lengths and types from six muscles of adult mice: three of them already described (flexor digitorum brevis, extensor digitorum longus, soleus) and three of them successfully dissociated for the first time (extensor hallucis longus, peroneus longus, peroneus digiti quarti).

Zebrafish are an emerging system for modeling human disorders of the skeletal muscle. We describe a fast and efficient method to isolate skeletal muscle myofibers from embryonic and larval zebrafish. This method yields a high-density myofiber preparation suitable for study of single skeletal muscle fiber morphology, protein subcellular localization, and muscle physiology.
