Wound Infection

Wound infection is the invasion and multiplication of microorganisms in damaged tissue, disrupting healing and potentially causing local or systemic illness. Bacteria and other pathogens can enter through a break in the skin, adhere to tissue, form biofilms, and trigger inflammation as immune cells and signaling molecules respond to the infection. This response may produce redness, warmth, swelling, pain, pus, delayed closure, or tissue damage, although symptoms can vary with the organism and wound type. Studying wound infection in biology supports research on host–pathogen interactions, antimicrobial treatment, immune defense, wound care, and strategies that reduce complications such as tissue loss or systemic infection.

Swine Model of Biofilm Infection and Invisible Wounds

2023

Chronic wounds that are resistant to antibiotics are a major threat to the healthcare system. Biofilm infections are stubborn and hostile and can cause deficient functional wound closure. We report a clinically relevant swine model of biofilm-infected full-thickness chronic wounds. This model is powerful for mechanistic studies as well as for testing interventions.

Establishment of Rice Plant Leaf Infection through Fungal Inoculation via Wounding

2025

This video demonstrates the establishment of rice plant leaf infection through wounding inoculation with Magnaporthe grisea mycelium. The fungal spore-derived germ tube penetrates the epidermal layer, resulting in tissue infection and lesions on the leaves. This method aids pathotype screening and improves plant resistance understanding for molecular breeding.

Bioluminescence Imaging of Acinetobacter baumannii-Infected Burn Wounds in Mice

Source: Wang, Y., et al. In Vivo Investigation of Antimicrobial Blue Light Therapy for Multidrug-resistant Acinetobacter baumannii Burn Infections Using Bioluminescence Imaging. J. Vis. Exp. (2017)This video demonstrates the procedure for imaging burn wound infections in mice inoculated with Acinetobacter baumannii carrying a bioluminescent operon. It demonstrates how luciferase-catalyzed substrate oxidation produces light proportional to bacterial load, enabling real-time monitoring of...

Generating an Ex Vivo Ovine Wounded Skin Model Infected with a Bacterial Pathogen

2025

The video demonstrates the generation of an ex vivo ovine skin model infected with Staphylococcus aureus. This entails treating a lamb's forehead skin biopsy section with antibiotics, establishing a wound bed, positioning it on a permeable membrane in a multiwell plate with antibiotic-free medium, and establishing bacterial infection in the skin wound.

Developing a Mouse Model of Chronic Pseudomonas aeruginosa Skin Wound Infection

Source: de Vries, C. R., et.al. A Delayed Inoculation Model of Chronic Pseudomonas aeruginosa Wound Infection. J. Vis. Exp. (2020)This video demonstrates the delayed inoculation of luminescent Pseudomonas aeruginosa into a mouse bearing two full-thickness dorsal skin wounds covered by a transparent film dressing. This facilitates biofilm formation and the establishment of chronic infection. Regular bioluminescence monitoring enables quantification of bacterial burden and tracking of infection...
