Spheroid Microplate

A spheroid microplate is a specialized culture plate that supports the formation and maintenance of three-dimensional cell aggregates, offering a more tissue-like model for cancer research than conventional two-dimensional cultures. Its nonadherent, often round-bottom wells promote cell-cell adhesion and aggregation, while standardized well geometry helps generate relatively uniform spheroids under controlled culture conditions. These models can reproduce important features of solid tumors, including cellular interactions, nutrient and oxygen gradients, and reduced drug penetration toward the spheroid core. Spheroid microplates therefore support tumor biology studies, therapeutic screening, and evaluation of treatment responses in physiologically relevant in vitro systems.

Introduction to the Microplate Reader

2023

The microplate reader is a multimodal instrument that allows for a variety of experiments to be performed and measured simultaneously. Microplate readers can make absorbance, fluorescence and luminescence measurements. Multiwell plates are integral to the microplate reader and allow for many experiments to be performed at once. Regardless of the assay type, experiments on the plate reader utilize a standard curve to determine the experimental values. This curve uses samples of known...

Generation of a Three-Dimensional Spheroid Model of Tumor Cell Invasion

2025

Source: Guyon, J., et al. A 3D Spheroid Model for Glioblastoma. J. Vis. Exp. (2020)The video demonstrates the generation of a three-dimensional spheroid model of tumor cell invasion. Spheroids are formed from human brain tumor cells and embedded in a collagen matrix. The tumor cells at the spheroid periphery adopt an invasive phenotype, secreting proteases to degrade the collagen and advancing to invade the surrounding matrix.

Splitting Spheroids for Sub-culturing and Shipping: A Procedure for Propagating and Transferring Spheroids from Small Bowel Neuroendocrine Tumor
