Thp-1 Differentiation

THP-1 differentiation is the laboratory conversion of THP-1 human monocytic leukemia cells into macrophage-like cells, providing a reproducible model for studying immune behavior in cancer. Typically, phorbol 12-myristate 13-acetate (PMA) activates protein kinase C signaling, causing the cells to adhere, change morphology, and acquire macrophage-associated functions and markers. Researchers use differentiated THP-1 cells to investigate tumor-associated macrophage biology, inflammatory signaling, cancer cell interactions, and responses to therapeutic compounds. Because the model is relatively accessible and standardized, it supports mechanistic studies and preliminary drug-screening approaches, although results require careful interpretation when extrapolated to primary human macrophages.

This protocol is based on LPS and ATP-induced death of PMA-differentiated THP-1 macrophages. We use flow cytometry to analyze Annexin V and 7-AAD double staining to detect cell death, using the whole cell and employing scanning electron microscopy to observe the cell membrane morphology.
