Heat-wait-seek Method

The Heat-wait-seek Method is a temperature-controlled nuclear magnetic resonance approach for studying thermally activated chemical reactions and molecular transformations over time. In a typical experiment, the sample is rapidly heated to initiate or accelerate a process, held for a defined waiting period, and then analyzed during a measurement step that captures changes in chemical species or local environments. By repeating this sequence with different temperatures or wait times, researchers can follow reaction kinetics, identify transient states, and evaluate temperature-dependent behavior. In chemistry, the method supports mechanistic studies of reactions, phase transformations, and dynamic processes that are difficult to resolve under steady-state conditions.

In this protocol we describe a method of operant learning using sensory stimuli as a reinforcer in the mouse. It requires no prior training or food restriction, and it allows the study of motivated behavior without the use of a pharmacological or natural reinforcer such as food.

A procedure that allows a demonstration of robust augmentation of drug seeking in food-restricted rats is described. Following heroin self-administration training, rats go through an abstinence period, in a drug-free environment, during which they are mildly food restricted. Drug seeking is then tested in the drug-associated environment.

Animal models of relapse, known as reinstatement procedures, have been used extensively to study the role of stress in relapse to drug seeking.
