Light Absorbance

Light absorbance is a dimensionless measure of how much incident light a sample removes at a selected wavelength, making it a central concept in chemical analysis. As light passes through a solution, molecules absorb photons at energies matching electronic, vibrational, or rotational transitions; under suitable conditions, the Beer-Lambert law relates absorbance to concentration and optical path length. Chemists use spectrophotometry to identify substances, quantify analytes, monitor reaction kinetics, and assess sample composition, while wavelength selection and calibration help produce reliable measurements in research, teaching, and quality control.

Here we introduce a method for using an intra-ventricle optical catheter in perfused hearts to perform absorbance spectroscopy across the heart wall. The data obtained provides robust information on tissue oxygen tension as well as substrate utilization and membrane potential simultaneously with cardiac performance measures in this ubiquitous preparation.
