Forebrain Electroporation

Forebrain electroporation is a gene-delivery technique that introduces nucleic acids into developing forebrain cells to investigate how genes regulate neural development. In this method, DNA or other genetic material is placed near embryonic brain tissue, and brief electrical pulses transiently increase membrane permeability, allowing the material to enter targeted cells; tissue polarity and electrode placement help determine the affected region. Researchers use the approach to manipulate gene expression in vivo and examine cell proliferation, migration, differentiation, axon formation, and circuit development. Because it is rapid and spatially targeted, forebrain electroporation supports functional studies of developmental mechanisms and disease-associated pathways.

Forebrain Electrophysiological Recording in Larval Zebrafish

A simple method to record extracellular field potentials in the larval zebrafish forebrain is described. The method provides a robust in vivo read-out of seizure-like activity. This technique can be used with genetically modified zebrafish larvae carrying epilepsy-related genes or seizures evoked by administration of convulsant drugs.

The present video demonstrates a method which takes advantage of the combination of electroporation and confocal microscopy to perform live imaging on individual neural progenitor cells in the developing zebrafish forebrain. In vivo analysis of the development of forebrain neural progenitor cells at a clonal level can be achieved in this way.
