Single Oocyte Assay

A single oocyte assay is an experimental method that measures the molecular, cellular, or functional properties of one egg cell, preserving differences that population-level analyses can obscure. Researchers isolate an individual oocyte, maintain it under defined conditions, and quantify responses such as maturation, activation, calcium signaling, gene expression, protein abundance, or metabolic activity using imaging or molecular readouts. In developmental biology, these assays help link oocyte quality and developmental competence to fertilization and early embryo development. They also support studies of meiotic regulation, maternal factors, reproductive disorders, and variability among oocytes, improving the precision of models for early developmental processes.

Bisulfite mutagenesis is the gold standard for analyzing DNA methylation. Our modified protocol allows for DNA methylation analysis at the single-cell level and was specifically designed for individual oocytes. It can also be used for cleavage-stage embryos.

In this video, we demonstrate that in the presence of a phosphodiester inhibitor, the oocyte cell cycle is arrested at prophase-I, during which transcribed mRNAs are stored in the absence of translation. After the release of the inhibitor, oocyte maturation resumes, and the cells enter metaphase. The signal of reporter mRNA fused with the target gene in metaphase-I indicates the activation of translational machinery and protein expression.

We describe a Xenopus oocyte and animal cap system for the expression cloning of genes capable of inducing a response in competent ectoderm, and discuss techniques for the subsequent analysis of such genes. This system is useful in the functional identification of a wide range of gene products.
