Liquid Cell Thickness

Liquid cell thickness is the distance across the confined liquid layer in a sealed cell, a key design parameter for imaging and analysis under liquid conditions. In electron microscopy, electrons pass through entrance and exit membranes plus the liquid, so increasing the liquid path length increases scattering and absorption, reducing transmitted signal and image contrast. Engineering the thickness therefore requires balancing sufficient volume for realistic reactions with the thin geometry needed for spatial resolution and manageable radiation exposure. Controlled liquid cell thickness supports in situ studies of nanoparticle growth, electrochemical processes, corrosion, and other dynamic phenomena that cannot be observed in a dry specimen.

This study presents a methodology to prepare 3D, biodegradable, foam-like cell scaffolds based on biocompatible side-chain liquid crystal elastomers (LCEs). Confocal microscopy experiments show that foam-like LCEs allow for cell attachment, proliferation, and the spontaneous alignment of C2C12s myoblasts.

We have developed a self-contained liquid cell, which allows imaging through liquids using a transmission electron microscope. Dynamic processes of nanoparticles in liquids can be revealed in real time with sub-nanometer resolution.

Organotypic Culture of Full-thickness Adult Porcine Retina

Here we describe a cost-effective technique for organotypic culture of adult porcine retina for seven days. Briefly, a sterile filter paper was used to lift the neural retina off from the RPE and place photoreceptor side up on an insert raised by a custom-made stand.
