Phloem Electrophysiology

Phloem electrophysiology is the study of electrical signals and membrane activity in phloem tissues, which transport sugars and signaling molecules through plants. In sieve elements and companion cells, ion movement across membranes generates changes in membrane potential that can propagate through the vascular network and interact with phloem transport. Environmental conditions such as wounding, temperature shifts, light changes, and water stress can influence these electrical responses, linking external stimuli to physiological regulation. Studying phloem electrophysiology helps explain long-distance plant communication, stress responses, resource allocation, and coordination between local tissues and distant organs.

Electrical Penetration Graph (EPG) is a well-established technique for studying the feeding behavior of stylet-bearing insects. Here we show a new application of EPG as a non-invasive tool for the acquisition of intracellular electrophysiology recordings of sieve elements (SEs), the cells that form the phloem vasculature in plants.

Knowledge of the composition of the phloem sap as well as the mechanism of its loading and long-distance transport is essential for the understanding of long-distance signaling in plant development and stress/pathogen response and of assimilate transport.
