Baculovirus Purification

Baculovirus purification is the process of separating baculovirus particles from infected cell cultures and contaminants to produce a concentrated, high-quality preparation for research and biotechnology. It typically combines clarification to remove cells and debris with filtration, centrifugation, density-gradient separation, or chromatography, using differences in particle size, density, or surface properties to isolate virions while preserving infectivity. In genetics, purified baculovirus serves as a reliable vector for delivering recombinant genes into insect or mammalian cells, supporting protein expression, gene-function studies, vaccine development, and gene delivery research. Purity and viral activity directly affect experimental reproducibility, yield, and downstream applications.

Heparin Affinity Chromatography-Based Baculovirus Purification: A Technique to Isolate Baculovirus From Insect Cell Supernatant

2025

In this video, we demonstrate the technique to isolate baculovirus from an insect cell supernatant using heparin affinity chromatography. The isolation is facilitated by the affinity between the heparin in the column to the baculovirus envelope glycoprotein.

In this protocol, baculovirus is produced by transient transfection of baculovirus plasmid into Sf9 cells and amplified in a serum-free suspension culture. The supernatant is purified by heparin affinity chromatography and further concentrated by ultracentrifugation. This protocol is useful for the production and purification of baculovirus for gene therapy application.

Generation of Chikungunya Virus-Like Particles Using Baculovirus Expression System in Insect Cells

This video shows how virus-like particles, VLPs are generated by infecting insect cells with a recombinant baculovirus carrying the chikungunya virus gene cassette. The recombinant virus infects cells, leading to VLP formation, assembly, and release into the culture medium for potential use in vaccine production.

In this protocol, AAV2 vector is produced by co-culturing Spodoptera frugiperda (Sf9) insect cells with baculovirus (BV)-AAV2-green fluorescent protein (GFP) or therapeutic gene and BV-AAV2-rep-cap infected Sf9 cells in suspension culture. AAV particles are released from the cells using detergent, clarified, purified by affinity column chromatography, and concentrated by tangential flow filtration.

Plasmid Purification

2023

Plasmid purification is a technique used to isolate and purify plasmid DNA from genomic DNA, proteins, ribosomes, and the bacterial cell wall. A plasmid is a small, circular, double-stranded DNA that is used as a carrier of specific DNA molecules. When introduced into a host organism via transformation, a plasmid will be replicated, creating numerous copies of the DNA fragment under study. In this video, a step-by-step generalized procedure is described for how to perform plasmid purification.
