Template Amplification

Template amplification is the process of generating many copies of a specific nucleic acid sequence from a small amount of starting material, making genetic information easier to detect and analyze. In polymerase chain reaction, primers define the target region, repeated heating separates DNA strands, primers anneal to complementary sequences, and a DNA polymerase extends them during cooling, producing exponential amplification. This approach supports genotyping, mutation detection, DNA sequencing, cloning, and analysis of samples with limited genetic material. By increasing target abundance while preserving sequence information, template amplification enables sensitive and reproducible investigations in genetics and molecular biology.

The study describes a protocol for creating large (µg-mg) quantities of DNA for protein screening campaigns from synthetic gene fragments without cloning or using living cells. The minimal template is enzymatically digested and circularized and then amplified using isothermal rolling circle amplification. Cell-free expression reactions could be performed with the unpurified product.

(2020).This video demonstrates the loop-mediated isothermal amplification (LAMP) technique for detecting a target bacterial gene by amplifying DNA at a constant temperature. The reaction mixture is prepared by adding an isothermal master mix, specific primers for the target gene, and samples containing DNA templates. The...

DNA as a Genetic Template

Two structural features of the DNA molecule provide a basis for the mechanisms of heredity: the four nucleotide bases and its double-stranded nature. The Watson-Crick model of double-helical DNA structure, proposed in 1952, drew heavily upon the X-ray crystallography work of researchers Rosalind Franklin and Maurice Wilkins. Watson, Crick, and Wilkins jointly received the Nobel Prize in Physiology or Medicine for their work in 1962. Franklin was, controversially, excluded from the prize for...
