Amoeba Lawn

Amoeba lawn is a microbiological assay in which free-living amoebae are spread across a layer of bacteria to model cellular predation and host-pathogen interactions. As amoebae move through the bacterial lawn, they engulf susceptible cells by phagocytosis and digest them, producing visible clear zones or plaques; bacterial survival indicates resistance to grazing or intracellular killing. In immunology and infection research, this method provides a simple, low-cost way to compare bacterial virulence, surface defenses, motility, and stress responses. Because amoebae share conserved features of phagocytic feeding with immune cells, the assay can help identify traits that influence bacterial persistence and guide further infection studies.

This article describes a simple, low-cost method of recording the lawn avoidance behavior of Caenorhabditis elegans, using readily available items such as a smartphone and a light emitting diode (LED) light box. We also provide a Python script to process the video file into a format more amenable for counting.
