Beta-hexosaminidase Release

Beta-hexosaminidase release is the secretion of the lysosomal enzyme beta-hexosaminidase from activated immune cells and serves as a measurable indicator of cellular degranulation. During activation, signaling pathways trigger cytoplasmic granules to fuse with the plasma membrane, releasing beta-hexosaminidase into the surrounding medium, where enzyme activity can be quantified with colorimetric or fluorogenic substrates. In immunology and infection research, this assay helps evaluate mast cell and basophil responses to allergens, pathogens, immune mediators, or candidate drugs. Results provide a practical measure of secretory activity and support studies of inflammation, host-pathogen interactions, and therapeutic inhibition of excessive immune activation.

Fabrication of Amyloid-Beta-Releasing Polymer Microbeads for Modeling Alzheimer's Disease

2025

This video demonstrates a method for fabricating polymer microbeads that encapsulate neurotoxic amyloid-beta-secreting cells. The cell-polymer suspension is processed through a microbead fabrication device, with polymer gelation achieved via calcium cross-linking, forming spherical microbeads around the cells. The polymer allows cell proliferation while allowing controlled, long-term amyloid-beta release, simulating its accumulation in Alzheimer's disease.

Humanized Mediator Enzyme Release Assay: An Immunochemical Method to Screen Allergic Potential of Test Antigen by Monitoring Basophil Degranulation Response In Vitro

2025

In this video, we describe an in vitro method to evaluate the allergic potential of an antigen to induce basophil degranulation. β-hexosaminidase is used as an index of chemical mediator release, and the enzyme activity of β-hexosaminidase is measured using a fluorogenic substrate.

Beta

2024

Beta is a crucial metric in finance used to assess an investment's risk and expected return relative to the overall market. It is calculated as the slope of the line in a scatterplot that compares the returns of an individual stock to the returns of the market benchmark, typically represented by a broad market index. Beta quantifies how much a stock's price moves in response to market movements, providing insight into its volatility and systematic risk. A beta of one indicates that the stock's...

Methods to Assess Beta Cell Death Mediated by Cytotoxic T Lymphocytes

Cell-mediated lymphocytotoxicity (CML) assays can be used to test autoreactive responses and study mechanisms of cell death in vitro. However, using live-cell confocal microscopic imaging techniques with fluorescent dyes, the type and kinetics of cell death as well as the pathways utilized can be studied in greater detail.

Imaging Transgenic Beta-Cells in an Immunodeficient Mouse Challenged with Diabetic Splenocytes

This video demonstrates an imaging-based method to visualize transplanted luciferase-expressing beta cells into a non-obese diabetic immunodeficient mouse. The introduction of splenocytes from a diabetic mouse leads to progressive beta cell destruction, visually confirmed by declining luminescence after substrate introduction.
