Blue-white Selection

Blue-white selection is a molecular cloning screening method that distinguishes bacterial colonies carrying recombinant DNA from those containing an empty vector. It relies on disruption of the lacZα gene: cells with an intact vector produce β-galactosidase, which cleaves X-gal and turns colonies blue, whereas insertion of foreign DNA interrupts lacZα complementation and produces white colonies. Researchers can then analyze white colonies by restriction digestion, PCR, or sequencing to confirm the insert. In immunology and infection research, this approach supports cloning genes encoding antigens, antibodies, virulence factors, and immune signaling proteins for expression, functional studies, and assay development.

Here we present methodology for the production of a focal stroke in murine white matter by local injection of an irreversible endothelial nitric oxide synthase (eNOS) inhibitor (L-Nio). The preparation and exfoliation of CaCuSi4O10 and BaCuSi4O10 are described. Upon stirring in hot water, CaCuSi4O10 spontaneously exfoliates into monolayers, whereas BaCuSi4O10 requires ultrasonication in organic solvents. Near infrared (NIR) imaging illustrates the NIR emission properties of these materials, and aqueous dispersions of these nanomaterials are useful for solution processing.
