Fc Antibody Blocking

Fc antibody blocking is a technique that prevents antibodies from binding nonspecifically to Fc receptors on immune cells, improving the accuracy of immunological assays. Before staining or detection, cells are treated with excess immunoglobulin, purified Fc fragments, or an Fc receptor-blocking reagent that occupies these receptors and limits unintended antibody interactions through the Fc region. This step is particularly important in flow cytometry, immunofluorescence, and tissue-based assays involving macrophages, dendritic cells, B cells, or other Fc receptor-bearing cells. By reducing background signal and false-positive results, Fc antibody blocking strengthens the interpretation of immune-cell phenotyping and infection-related experiments.

We describe a method to measure the activation of Fc-mediated effector functions by antibodies that target the influenza virus hemagglutinin. This assay can also be adapted to assess the ability of monoclonal antibodies or polyclonal sera targeting other viral surface glycoproteins to induce Fc-mediated immunity.

In this study, we describe an improved protocol for a multiplexed high-throughput antibody microarray with lectin detection method that can be used in glycosylation profiling of specific proteins. This protocol features new reliable reagents and significantly reduces the time, cost, and lab equipment requirements as compared to the previous procedure.
