Neutrophil Biofilm Interaction

Neutrophil biofilm interaction describes how neutrophils, a major innate immune cell, recognize and respond to microbial communities encased in a protective extracellular matrix. Neutrophils migrate toward biofilms through chemotactic signals and deploy phagocytosis, reactive oxygen species, antimicrobial enzymes, and neutrophil extracellular traps, while the matrix and biofilm-associated factors can limit antimicrobial penetration and alter neutrophil function. Understanding this host-microbe interaction helps explain persistent inflammation and impaired clearance in chronic wounds, cystic fibrosis, and device-associated infections. It also supports research into therapies that improve neutrophil activity, disrupt biofilm structure, or combine immune modulation with antimicrobial treatment.

The present protocol describes the study of neutrophil-biofilm interactions. Staphylococcus aureus biofilms are established in vitro and incubated with peripheral blood-derived human neutrophils. The oxidative burst response from neutrophils is quantified, and the neutrophil localization within the biofilm is determined by microscopy.
