Optical Density

Optical density is a spectrophotometric measure of how strongly a sample attenuates light, providing an indirect estimate of its concentration or turbidity. In immunology and infection research, it is commonly assessed by directing light of a defined wavelength through a culture and recording absorbance or light scattering, often at 600 nm for bacterial suspensions. As microbial cells multiply, the suspension becomes more turbid and its optical density rises, allowing researchers to follow growth kinetics, standardize inocula, and compare antimicrobial or immune-mediated effects. These measurements support studies of pathogen burden, antimicrobial susceptibility, and host cell responses, while careful blanking, calibration, and consistent pathlength improve reproducibility.

An Adapted Optical Density-Based Microplate Assay for Characterizing Actinobacteriophage Infection

An optical density-based microplate method is described to quantify long-term bacterial growth in the presence of bacteriophages, suitable for actinomycetes and other slow-growing bacteria. This method includes modifications to reduce evaporation and lid condensation and R code to calculate virus infection metrics, including the area under the curve, growth maximum, and relative virulence.
