Thp-1 Macrophages

THP-1 macrophages are macrophage-like cells generated from the human THP-1 monocytic leukemia cell line and widely used to model innate immune responses in vitro. Treatment with phorbol 12-myristate 13-acetate (PMA) activates protein kinase C signaling, promoting monocyte adhesion, differentiation, and acquisition of macrophage-associated functions; subsequent resting and immune stimulation can further shape their response. In immunology and infection research, THP-1 macrophages support studies of pathogen recognition, phagocytosis, inflammatory cytokine production, host-pathogen interactions, and antimicrobial or anti-inflammatory compounds. Their standardized, scalable culture makes them useful for mechanistic experiments while complementing studies using primary human macrophages.

This protocol is based on LPS and ATP-induced death of PMA-differentiated THP-1 macrophages. We use flow cytometry to analyze Annexin V and 7-AAD double staining to detect cell death, using the whole cell and employing scanning electron microscopy to observe the cell membrane morphology.
