Drug Deposition Rate

Drug deposition rate is the speed at which a medication reaches and accumulates at its intended biological site, an important determinant of exposure, efficacy, and safety. In inhaled therapy, deposition occurs as drug-containing particles travel through the airways, where particle size, airflow, breathing pattern, formulation, and airway anatomy influence whether they impact the mouth, throat, lungs, or are exhaled. Measuring and controlling deposition rate helps researchers optimize drug delivery, compare formulations, predict local and systemic concentrations, and reduce off-target effects. This concept supports the development of inhaled treatments and other dosage forms that require precise spatial and temporal drug distribution.

Renal Drug Excretion: Effect of Urine pH, Flow Rate, and Drug pKa

2025

The pH of urine, the drug's pKa, and the urine flow rate are vital parameters for drug reabsorption and excretion. Urinary pH varies between 4.6 and 8.0 and is influenced by diet, drug intake, and the patient's pathophysiology. It affects a drug's ionization state and reabsorption. For instance, carbohydrate-rich food produces alkaline urine promoting drug excretion, while proteins and certain medications like ascorbic acid lead to acidic urine enhancing reabsorption. The pKa of a drug,...

Factors Affecting Drug Distribution: Organ Perfusion Rate

2025

Drug distribution within the body is a complex process influenced by several factors, including perfusion rate, the rate at which the bloodstream transports drugs to tissue. This limitation becomes particularly significant when dealing with highly lipophilic drugs. In such cases, the rate at which the drug can move across membranes is crucial, and if the membrane is highly permeable to the drug, distribution becomes rate-limited by perfusion. Perfusion rate-limited distribution relies on the...

Phase Transitions: Sublimation and Deposition

2020

Some solids can transition directly into the gaseous state, bypassing the liquid state, via a process known as sublimation. At room temperature and standard pressure, a piece of dry ice (solid CO2) sublimes, appearing to gradually disappear without ever forming any liquid. Snow and ice sublimate at temperatures below the melting point of water, a slow process that may be accelerated by winds and the reduced atmospheric pressures at high altitudes. When solid iodine is warmed, the solid sublimes...

Reaction Rate

The rate of reaction is the change in the amount of a reactant or product per unit time. Reaction rates are therefore determined by measuring the time dependence of some property that can be related to reactant or product amounts. Rates of reactions that consume or produce gaseous substances, for example, are conveniently determined by measuring changes in volume or pressure. The mathematical representation of the change in the concentration of reactants and products, over time, is the rate...
