Hepatic Regeneration

Hepatic regeneration is the liver’s ability to restore lost tissue and recover function after injury, making it a central process in medicine and liver disease research. Following partial hepatectomy or toxic damage, surviving hepatocytes re-enter the cell cycle in response to coordinated signals from cytokines, growth factors, and the extracellular matrix, while nonparenchymal cells support tissue remodeling. Regeneration can restore liver mass and metabolic capacity, although chronic injury may disrupt this response and promote fibrosis or cancer. Understanding these mechanisms informs treatments for liver failure, improves surgical planning, and supports research into transplantation, drug toxicity, and regenerative therapies.

Tools used for visualizing vascular regeneration require methods for contrasting the vascular trees. This film demonstrated a delicate injection technique used to achieve optimal contrasting of the vascular trees and illustrate the potential benefits resulting from a detailed analysis of the resulting specimen using µCT and histological serial sections.

An optical system was developed to visualize hepatic microcirculation with FITC-labeled erythrocytes and to measure the partial pressure of oxygen in the microvessels with laser-assisted phosphorimetry. This method can be used to investigate physiological and pathological mechanisms by analyzing microvascular structure, diameter, blood flow velocity, and oxygen tension.
