Interphase Fish

Interphase FISH (fluorescence in situ hybridization) is a cytogenetic method that detects specific DNA sequences in nondividing cells, providing rapid genetic information when metaphase chromosomes are unavailable. In fixed interphase nuclei, fluorescently labeled DNA probes bind complementary target sequences through nucleic-acid hybridization, and microscopy reveals the number, position, or pattern of signals. In medicine, this approach supports the detection of gene amplifications, deletions, rearrangements, and aneuploidy in tumor samples and other clinical specimens. Its speed and ability to analyze uncultured cells make it useful for diagnostic testing, treatment planning, and monitoring genomic changes in disease.

Here we describe a protocol for simultaneous detection of histone modifications by immunofluorescence and DNA sequences by DNA FISH followed by 3D microscopy and analyses (3D immuno-DNA FISH).
