Microfluidic Cartridge

A microfluidic cartridge is a compact device that directs and processes small volumes of fluids through networks of microscale channels, enabling controlled analysis with limited samples and reagents. Its channels use precisely designed geometries, surface properties, and fluid forces to transport, mix, separate, or react samples, often integrating valves, sensors, and detection zones within a disposable format. In medicine, microfluidic cartridges support point-of-care diagnostics, blood and cell analysis, nucleic acid testing, and drug-response studies. By combining sample preparation and measurement in one platform, they can shorten workflows, reduce material use, and facilitate rapid testing near patients or in resource-limited settings.

This video demonstrates a protocol for detecting serum-specific immunoglobulin E using a microfluidic cartridge-based chemiluminescence system. Serum IgEs interact with distinct allergens immobilized in the reaction zone of the cartridge, which are subsequently detected with peroxidase-conjugated anti-IgE antibodies and a chemiluminescent substrate, allowing simultaneous detection of multiple allergen-specific IgEs.

Use of a Filter Cartridge for Filtration of Water Samples and Extraction of Environmental DNA

We describe a protocol for filtration of water samples with a filter cartridge and extraction of environmental DNA (eDNA) without having to cut open the housing to remove the filter. This protocol is developed for metabarcoding eDNA from fishes, but is also applicable to eDNA from other organisms.

Multi-step Variable Height Photolithography for Valved Multilayer Microfluidic Devices

Multilayer microfluidic devices often involve the fabrication of master molds with complex geometries for functionality. This article presents a complete protocol for multi-step photolithography with valves and variable height features tunable to any application. As a demonstration, we fabricate a microfluidic droplet generator capable of producing hydrogel beads.

A Microfluidic Chip for ICPMS Sample Introduction

We present a discrete droplet sample introduction system for inductively coupled plasma mass spectrometry (ICPMS). It is based on a cheap and disposable microfluidic chip that generates highly monodisperse droplets in a size range of 40−60 µm at frequencies from 90 to 7,000 Hz.
