Respiratory Muscle Training

Respiratory muscle training is a structured exercise approach that strengthens the muscles responsible for breathing and can improve ventilatory function. It typically uses devices that impose resistance during inhalation or exhalation, creating a controlled workload that promotes adaptation in the diaphragm and other respiratory muscles. In medicine, clinicians use this training to support people with chronic respiratory disease, neuromuscular disorders, reduced exercise tolerance, or impaired breathing after illness or surgery. By improving respiratory muscle strength and endurance, the intervention may reduce breathlessness, enhance exercise capacity, and support functional recovery when integrated into an individualized rehabilitation program.

A murine model of neuromuscular electrical stimulation (NMES), a safe and inexpensive clinical modality, to the anterior compartment muscles is described. This model has the advantage of modifying a readily available clinical device for the purpose of eliciting targeted and specific muscle contractions in mice.

Here, we describe a protocol to record and analyze respiratory electromyography (EMG) signals. It includes the anatomic references for placing the EMG electrodes over several respiratory muscles, removing electrocardiographic noise from the EMG signals, and acquiring the EMG root mean square (RMS) and onset timing of activity.
