Bipolar Cell Terminals

Bipolar cell terminals are specialized presynaptic endings of bipolar neurons that transmit signals between upstream sensory cells and downstream neurons, making them essential for visual information processing in the retina. They convert graded changes in membrane potential into neurotransmitter release at ribbon synapses, where calcium entry regulates glutamate secretion onto retinal ganglion and amacrine cells. Distinct ON and OFF bipolar pathways respond differently to changes in light intensity, preserving contrast and temporal information as signals move through retinal circuits. Studying these terminals clarifies synaptic transmission, visual coding, and the cellular mechanisms underlying retinal disorders and emerging vision-restoration strategies.

Here, we present a method for light microscopy analysis of tracheal terminal cells in Drosophila larvae. This method allows for quick examination of branch and lumen morphology in whole animals and would be useful for analysis of individual mutants or screens for mutations affecting terminal cell development.
