Cac Knockdown Flies

Cac knockdown flies are Drosophila melanogaster engineered to reduce expression of cacophony (Cac), a gene encoding a presynaptic voltage-gated calcium channel essential for neuronal communication. In neurons, Cac channels open when an action potential reaches the synaptic terminal, allowing calcium influx that triggers synaptic vesicle fusion and neurotransmitter release; knockdown weakens this process by lowering channel abundance or function. These flies provide a genetic model for studying calcium-dependent synaptic transmission, active-zone organization, and neural circuit function. By linking altered Cac activity to changes in electrophysiology, behavior, or development, researchers can clarify mechanisms underlying neuronal signaling and synaptic disorders.

Drosophila has emerged as a significant model system for dissecting the cellular and molecular underpinnings of behavioral responses to alcohol. Contribution of the ACF chromatin remodeling factor to E4orf4-induced cell death was measured. The protocol includes selection of cell clones in which doxycycline treatment induces conditional knockdown of the ACF subunits Acf1 and SNF2h, and use of the DAPI assay to measure E4orf4-induced cell death in the inducible cell lines.
