Degenerative Loading Protocol

A degenerative loading protocol is a controlled experimental procedure that applies defined mechanical, metabolic, or physiological stress to model progressive loss of neural structure or function. By regulating the magnitude, duration, and repetition of the load, researchers can examine how neural tissue responds when stress exceeds its capacity for adaptation and recovery. In neuroscience, these protocols help standardize studies of degeneration, allowing investigators to compare cellular injury, changes in neural activity, tissue remodeling, and recovery across experimental conditions. They support research into disease mechanisms, injury progression, therapeutic efficacy, and biomarkers while improving the reproducibility of preclinical models.

In vitro use of degenerative tenocytes is essential when investigating the efficacy of novel treatment on tendinopathy. However, most research studies use only the animal model or a healthy tenocyte. We propose the following protocol to isolate human degenerative tenocytes during surgery.

Degenerative Disc Disease I: Introduction

2026

Degenerative disc disease is a chronic condition in which intervertebral discs gradually lose structure and function. It is not infectious or autoimmune; rather, it results from age-related biochemical and mechanical changes, influenced by genetic, metabolic, and environmental factors.Structure and Function of DiscsThe spine contains 23 intervertebral discs that absorb load, distribute forces, maintain spacing, and allow flexibility. Each disc consists of a nucleus pulposus, a gel-like core...

Degenerative Disc Disease ll: Pathophysiology

The symptoms of degenerative disc disease arise from a combination of mechanical compression, vascular compromise, and biochemical inflammation, which together disrupt nerve function and produce pain.Mechanical CompressionDisc degeneration reduces height and elasticity, predisposing to herniation of the nucleus pulposus, a major cause of radicular pain. Herniations may be protrusion (bulging with intact annulus), extrusion (nucleus extends beyond disc but remains connected), or sequestration...

Impact Loading

2024

Impact loading occurs when a moving object collides with a stationary structure, such as a rod with a uniform cross-sectional area fixed at one end. Under these conditions, the rod absorbs the kinetic energy from the striking object, leading to deformation and subsequent stress development. As the rod returns to its original position and reaches maximum stress, the absorbed energy, initially manifested as kinetic energy, transforms entirely into strain energy. In cases of elastic deformation,...

Eccentric Loading

Eccentric loading is a crucial concept in the study of structural engineering and mechanics, particularly when analyzing the stability and stress distribution in columns. Unlike centric loading, where the force is applied along the centroidal axis, causing uniform compression, eccentric loading occurs when a force is applied off-center. This off-center application introduces not only direct compressive stress but also bending stress, significantly influencing the column's behavior under load.
