Dipeptide Repeat Proteins

Dipeptide repeat proteins (DPRs) are repetitive polypeptides produced from expanded nucleotide sequences, especially the C9orf72 repeat expansion, and are important because they contribute to neurodegeneration. Through repeat-associated non-ATG translation, expanded repeat RNA can generate DPRs without a conventional start codon; these proteins accumulate in the nucleus and cytoplasm, interact with RNA-binding proteins and cellular transport machinery, and disrupt nucleocytoplasmic transport, RNA processing, and protein homeostasis. In neuroscience, studying DPR production, localization, and toxicity helps explain mechanisms underlying amyotrophic lateral sclerosis and frontotemporal dementia, while supporting the development of biomarkers and therapeutic strategies that target repeat RNA, translation, or DPR clearance.
