Immunoperoxidase Dab Staining

Immunoperoxidase DAB staining is an antibody-based histological technique that visualizes specific proteins in fixed tissue, making it valuable for mapping cellular and molecular features in neuroscience. Primary antibodies bind target antigens, while enzyme-linked secondary antibodies typically carry horseradish peroxidase; in the presence of hydrogen peroxide, the enzyme oxidizes 3,3′-diaminobenzidine (DAB) to form an insoluble brown precipitate at the antigen’s location. Researchers use this signal to localize neurotransmitters, receptors, neuronal markers, and glial proteins within brain sections. The method links molecular identity with tissue architecture and supports studies of neural circuits, development, disease, and injury.
