Intracardiac Perfusion Fixation

Intracardiac perfusion fixation is a tissue-preservation technique that uses the cardiovascular system to rapidly replace blood with a fixative, helping maintain the structure and molecular features of organs such as the brain. After anesthesia, a cannula delivers buffered saline through the left ventricle to clear circulating blood, followed by fixative that travels through cerebral vessels while fluid drains from the right side of the heart. In neuroscience, this method improves preservation of neural tissue for histology, immunohistochemistry, and microscopy, supporting accurate analysis of brain anatomy, cellular organization, and disease-related changes.

Air-Inflation of Murine Lungs with Vascular Perfusion-Fixation

Presented is a method for air-inflation with vascular perfusion-fixation of the lungs that preserves the location of cells within airways, alveoli and interstitium for structure-function analyses. Constant airway pressure is maintained with an air-inflation chamber while fixative is perfused via the right ventricle. Lungs are processed for histologic studies.

The porcine model of liver normothermic machine perfusion (NMP), described here, can be successfully used to study NMP as a preservation strategy, a tool for viability assessment, and a platform for organ repair. It holds a high translational value, however it is technically challenging and labor-intensive.
