Micromanipulator Control

Micromanipulator control is the precise positioning and movement of small instruments within a laboratory workspace, enabling researchers to handle cells, tissues, electrodes, and other delicate specimens with minimal disturbance. Mechanical knobs, motorized drives, or joystick interfaces convert user input into finely scaled movements along one or more axes, allowing a pipette or recording electrode to approach a target under microscopic visualization while maintaining stable placement. In neuroscience, micromanipulator control supports patch-clamp recording, intracellular stimulation, microinjection, and targeted manipulation of neurons or brain tissue. Accurate control improves recording quality, experimental reproducibility, and access to cellular mechanisms underlying neural signaling.

Microiontophoresis and Micromanipulation for Intravital Fluorescence Imaging of the Microcirculation

Microiontophoresis entails movement of ions from a micropipette in response to a difference in electrical potential between the inside and outside of the micropipette. Biologically active molecules are thereby delivered in proportion to electrical current. We illustrate acetylcholine microiontophoresis in conjunction with micromanipulation to study endothelium-dependent vasodilation in the microcirculation.

We describe how micro- and photomanipulation techniques such as FRAP and photoactivation enable the determination of motility parameters and the spatiotemporal dynamics of proteins within migrating cells. Experimental readouts include subcellular dynamics and turnover of motility regulators or of the underlying actin cytoskeleton.

Here, we present an integrated workflow to identify phenotypic and molecular features that characterize circulating tumor cells (CTCs). We combine live immunostaining and robotic micromanipulation of single and clustered CTCs with single cell-based techniques for downstream analysis and assessment of metastasis-seeding ability.
