Periplaneta Americana

Periplaneta americana, commonly called the American cockroach, is a resilient insect species used to study neural organization, sensory processing, and behavior. Its antennae, mechanosensory hairs, and other receptors detect environmental cues, sending signals through the ventral nerve cord to interneurons that coordinate rapid leg movements and escape responses. In neuroscience, this accessible nervous system supports investigations of synaptic transmission, sensory integration, motor control, and locomotor circuits. Because its behaviors are robust and its neural pathways can be examined experimentally, Periplaneta americana provides a valuable comparative model for understanding how nervous systems convert sensory information into coordinated action.

This protocol is used in establishing and maintaining gnotobiotic American cockroaches (Periplaneta americana) by surface sterilizing the egg cases (oothecae) prior to hatching. These gnotobiotic insects contain their vertically transmitted Blattabacterium endosymbionts but have axenic guts.

We demonstrate variations of the extracellular multi-unit recording technique to characterize odor-evoked responses in the first three stages of the invertebrate olfactory pathway. These techniques can easily be adapted to examine ensemble activity in other neural systems as well.
