Silicone Pin Mounting

Silicone pin mounting is a preparation technique that uses pins and a silicone elastomer support to stabilize biological specimens for manipulation, dissection, or imaging. The cured silicone provides a compliant, nonrigid surface, while strategically placed pins anchor the specimen and restrict movement without obstructing access to surrounding tissue or solution. In neuroscience, this approach helps maintain the position of neural tissues or model organisms during microscopic examination, tissue isolation, staining, and electrophysiological procedures. By improving stability and reproducibility, silicone pin mounting supports precise observations of nervous-system structure and function and can make delicate experimental workflows easier to perform.

We present a method to isolate the adult organ of Corti as three intact cochlear turns (apex, middle, and base). We also demonstrate a procedure for immunostaining with fluorescently tagged antibodies. Together these techniques allow the study of hair cells, supporting cells, and other cell types found in the cochlea.

Euler's Formula for Pin-Ended Columns

2024

In structural engineering, the stability of columns under compressive axial loads is a critical consideration, described as buckling. A typical example involves a column PQ, which is pin-connected at both ends and subjected to a centric axial load F applied at one end, with a reaction force of F' = -F at the other end. Here, it is crucial to understand that when an applied load exceeds the critical load, buckling occurs as the system becomes unstable. To calculate the critical load, envision...

Large-scale Recording of Neurons by Movable Silicon Probes in Behaving Rodents

We describe methods for large-scale recording of multiple single units and local field potential in behaving rodents with silicon probes. Drive fabrication, probe attachment to the drive and probe implantation processes are illustrated in sufficient details for easy replication.

Construction Methods for Hybrid Polycarbonate-Silicon Microfluidic Devices

2026

This protocol describes three straightforward sealing methods for fabricating a hybrid microfluidic device that integrates polycarbonate and silicon components. By implementing solvent bonding, thermal compression, and mechanical compression strategies, we enable reliable, low-temperature sealing techniques that avoid specialized equipment for hybrid microfluidic devices.
