Cation Flux

Cation flux is the movement of positively charged ions, such as sodium, potassium, calcium, and protons, across cell membranes, a process that supports electrical signaling and cellular regulation. It occurs when ion channels open or transporters operate, allowing cations to move down or against electrochemical gradients established by membrane potential and active pumps. In pharmacology, measuring or modifying cation flux helps explain how drugs influence neuronal transmission, muscle contraction, cardiac rhythm, secretion, and cell survival. Ion channel blockers, activators, and transporter inhibitors can therefore reveal drug mechanisms and provide therapeutic strategies for neurological, cardiovascular, and other diseases.

Measuring Cation Transport by Na,K- and H,K-ATPase in Xenopus Oocytes by Atomic Absorption Spectrophotometry: An Alternative to Radioisotope Assays

We describe a method to quantify the activity of K+-countertransporting P-type ATPases by heterologous expression of the enzymes in Xenopus oocytes and measuring Rb+ or Li+ uptake into individual cells by atomic absorption spectrophotometry. The method is a sensitive and safe alternative to radioisotope flux experiments facilitating complex kinetic studies.

Transporters in cell membranes allow differential segregation of ions across cell membranes or cell layers and play crucial roles during tissue physiology, repair and pathology. We describe the ion-selective self-referencing microelectrode that allows the measurement of specific ion fluxes at single cells and tissues in vivo.
