Drug Synergism

Drug synergism is a pharmacological interaction in which the combined effect of two or more drugs exceeds the effect expected from their individual activities, making it important for understanding combination therapy. Synergy can occur when agents act on complementary molecular targets or biological pathways, enhance one another’s absorption or activity, or block compensatory responses that limit treatment. Pharmacologists evaluate these interactions by comparing observed combination effects with predicted additive effects under defined conditions. This analysis supports the design of more effective therapies, potentially allowing lower doses and reduced toxicity, while also helping identify harmful interactions and guide treatment strategies for complex diseases.

Synergism is a useful mechanism where combining two or more drugs is more effective than each constituent used alone. Such combinations are also called supra-additive interactions. The drugs collectively enhance the final therapeutic effect by acting on different targets. A validated computerized battery of neuropsychological and neurophysiological tests is used to study pharmacodynamic effects on the central nervous system of newly developed drugs in early phase development. To demonstrate the test battery, the acute effects of mecamylamine and the reversal of these effects by two agonist drugs are described.
