Hepatic Metabolism

Hepatic metabolism is the enzymatic transformation of drugs and other foreign compounds in the liver, helping regulate their activity, duration, and elimination from the body. Hepatocytes commonly carry out Phase I reactions, such as oxidation by cytochrome P450 enzymes, followed by Phase II conjugation, which attaches polar groups to increase water solubility and support excretion in urine or bile. In pharmacology, these processes influence oral bioavailability through first-pass metabolism, determine active or toxic metabolites, and contribute to drug interactions when compounds inhibit or induce metabolic enzymes. Understanding hepatic metabolism supports safer dose selection and more predictable therapeutic outcomes.

A progressive colonization procedure is described to further assess its impact on the host hepatic metabolism. Colonization is monitored non invasively by evaluating the urinary excretion of microbial co-metabolites by NMR-based metabolic profiling while hepatic metabolism is assessed by High Resolution Magic Angle Spinning (HR MAS) NMR profiling of intact biopsy.
